Principle:- Chromogenic Coliform Agar is a selective medium recommended for the simultaneous detection and recovery of sub-lethally injured coliforms in water samples. The CCA media is composed of tryptone, yeast extract, sodium chloride, sodium dihydrogen phosphate, disodium hydrogen phosphate, sodium pyruvate, sorbitol, tryptophan, tergitol-7 and three chromogenic substrates. The three chromogenic substrates are 6- chloro-3-indoxyl β-D-galactopyranoside, 5-bromo-4-chloro-3-indoxyl- β-D-glucuronic acid cyclohexamine ammonium salt, monohydrate and IPTG (Isopropyl-β-D-thiogalactopyranoside). Tryptone, sodium pyruvate and sorbitol provide nitrogenous substances, fermentable carbohydrate and other essential growth nutrients for the organisms. The L-Tryptophan improves the indole reaction thus increasing the revealing reliability.
Use:- Recommended for detection of Escherichia coli and coliforms in water samples. The composition and performance criteria of this medium are as per the provisions laid down in ISO 9308-1:2014.

• Principle:- The Tryptone serves the source of nitrogen, yeast extract serves the source of nitrogen and vitamins, and sodium chloride balances the osmotic balance. The Luria Bertani (Miller) is used for the cultivation of E.coli. The robust growth of E. coli is necessary for the cultivation of microphages and in the recombinant technology studies.
• Use:- Recommended For cultivation of recombinant strains of E. coli.
• Directions:- Suspend 40.0 gram in 1000 ml distilled water adjust the pH if required. Boil to dissolve the medium completely. Luria Bertani Broth sterilize by autoclaving at 15 lbs pressure (121 °C) for 15 min.
• Performance and Evaluation:- The expected performance of the medium is liable to use as per the direction on the label when stored at optimum conditions and within expiry date.
• Storage and Shelf Life:- Hygroscopic; keep container tightly closed. Store in cool dry place.
