• MacConkey Broth , which contains neutral red as an indicator is considered as a standard medium for the primary isolation as well as presumptive identification of coliform-aerogenes group of organisms in food and water.
• Principle:- Peptone serves as the source of all essential nutrients such as amino acids, vitamins, other trace factors which are essential for the growth of microbes. The lactose in it plays a crucial role for selection of lactose fermenting microbes. Moreover the combination of Bromo Cresol Purple and the bile salts inhibits the Gram-positive bacteria.
• Use:- Recommended for selective isolation and differentiation of Lactose fermenting and Lactose non-fermenting enteric bacteria from water, sewage etc. Specifically the coliforms; E coli by using Durham’s tube for detection of gas production. The MacConkey Broth is recommended for the use in microbial examination of water samples, milk and dairy products, and recommended by many regulatory bodies such as British Pharmacopeia.
• Directions:- Suspend 40 gram in 1000 ml distilled water adjust the pH if required. MacConkey Broth sterilize by autoclaving at 15 lbs pressure (121 °C) for 15 min, cool it to 42-45 °C.

• Principle:- Malt Extract Powder is prepared from aqueous extract of sprouted malt grains and dried at low temperature to preserve nutrients present in the form of carbohydrates and nitrogenous substances.

Use : Recommended for identificaton of gram-negative enteric bacilli on the basis of dextrose, lactose and sucrose fermentation and hydrogen sulphide production.

Directions : Dissolve 17.00 Grams in 1000 ml distilled water. Boil to dissolve the medium completely and distribute aseptically. Sterilize by autoclaving at 15 lbs pressure (121 'C) for 15 min, kept in slight slanting position to form small slant with large butt. Ensure complete solidification and inoculate test sample aseptically.

Principle:- Sabouraud dextrose broth is used for cultivation of yeasts, molds and aciduric bacteria. It is recommended by USP as a medium for microbial limit testing of pharmaceutical products and raw material used in pharmaceutical industries. The medium is prepared in accordance with the harmonized principles of USP. Medium is consisting of meat and casein peptone (1:1) and dextrose monohydrate. The meat and casein peptone provide carbonaceous, nitrogenous compounds, long chain amino acids, vitamins and other essential growth nutrients in addition to that the dextrose monohydrate serves as energy source. The high concentration of dextrose and low pH of medium favor the growth of yeasts and molds and inhibit other contaminating bacteria from pharmaceutical and clinical specimens. In addition to pH the Chloramphenicol provides excess selective for the growth of fungi and yeast.

Use:- Recommended for the cultivation of yeasts and molds from pharmaceutical products in accordance with the microbial limit testing by harmonized principles of USP.
