Principle:-
Agar is a Phycocolloid extracted from the red-purple marine algal group (class Rhodophyceae) including GelediumPterocladia and Glcilaria. The agar is used as the gelling agent as it remained the firm at the incubation temperatures of bacteria and could not be utilized by microbes. Principle:- Asparagine proline broth is an enrichment broth for Pseudomonas aeruginosa. The composition is strictly mineral base with enantiomeric forms of asparagine and proline, magnesium sulphate, dipotassium sulphate and potassium sulphate. DL-asparagine and proline serve as the sole source of nitrogen. The dipotassium and potassium salts act as a buffer system and magnesium sulfate perform multiple functions, magnesium ion required in a large variety of enzymatic reactions, reproduction and also acts as a buffer. Pseudomonas aeruginosa hydrolyze asparagine to aspartic acid and reproduce vigorously, the appearance of growth with or without fluorescent pigmentation is considered a presumptive test for P. aeruginosa. The microbial count can be determined by using the MPN test and conformation need to be done by subculturing on cetrimide agar. Precautions to be taken:-  Asparagine Proline Broth these microbial media are intended for the in-vitro use only. All the handling, experiments, storage, and discarding should be performed with the help of skilled and knowledgeable technicians and as per the established guidelines. The material should be disposed only after proper sterilization by autoclaving. Please go through the MSDS of the media to avoid any accidents or in emergency. Directions:- Dissolve 14.50 grams in 900 ml distilled water, boil to dissolve the medium completely allow to cool at room temperature and add 25ml of ethanol and dilute up to 1000 ml with distilled water. Distribute as desired in screw-capped bottles. Do not used polypropylene caps with seat. Close the caps so that the seal in the lid just touches the lip of the bottle. Sterilize by autoclaving at 15 lbs pressure (121 °C) for 15 min, cool it to 42-45 °C and inoculate test sample aseptically. Alternatively, sterilize the ethanol by filtration through the cellulose acetate or nitrate membrane of 0.22 µm and then aseptically to the medium after autoclaving and cooling.
Use:- Recommended for the cultivation of Pseudomonas aeruginosa using membrane filter technique.

Potato Dextrose Broth is recommended for the isolation and enumeration of yeasts and moulds.
Principle:- Potato infusion, serve as the source of all essential nutrients such as amino acids, vitamins, other trace factors which are essential for the growth of microbes and dextrose in it supply the C source and energy source fermenting microbes.Moreover the potato infusion encourages the sporulation and pigment production in some dermatophytes. The potato dextrose Broth can be supplemented with the acid to reduce pH, or the antibiotics for inhibition of bacterial growth.Use:- Recommended for the cultivation of yeasts, and moulds from clinical and non-clinical samples. Also, recommended for the plate count methods for the food, dairy products.
Directions:- Suspend 24.0 gram in 1000 ml distilled water adjust the pH if required. Boil to dissolve the medium completely. Sterilize by autoclaving at 15 lbs pressure (121 °C) for 15 min. After autoclaving the pH of the medium can be set to 3.5 ±0.2 by addition of the 10% Tartaric acid (Quantity is specified on each container).

Principle

  • EMB agar is characterized by the presence of a combination of the two dyes eosin and methylene blue in the ratio of 6:1.
  • Gram-negative bacteria that ferment the lactose produce acid which lowers the pH. This encourages dye absorption by the colonies and turns the colonies dark purple as the acid acts upon the dyes.
  • In addition, certain lactose-fermenting bacteria produce flat, dark colonies with a green metallic sheen.
  • Other lactose fermenters produce larger, mucoid colonies, often purple only in their center.
  • In EMB agar, most of the strains of  E. coli colonies have a characteristic green sheen. Rapid fermentation of lactose & production of strong acids, thus a rapid reduction in the pH of the EMB agar the critical factor in the formation of the green metallic sheen observed with E. coli, rapid fermentation of lactose and formation of strong acids.

Principle And Interpretation 

Baird Parker Agar was developed by Baird Parker  from the Tellurite-glycine formulation of Zebovitz et al  for isolation and enumeration of Staphylococci in food and other material since it allows a good differentiation of coagulase positive strains. A high correlation has been found between the coagulase test and the presence of clear zone of lipolysis in this medium, which is due to the lecithinase of Staphylococci that breakdown, the egg yolk. On the other hand, studies show that almost 100% of coagulase positive Staphylococci are capable of reducing tellurite, which produces black colonies, whereas other Staphylococci cannot always do so.

CLED agar (cystine lactose electrolyte deficient agar) is a valuable non-inhibitory  growth medium used in the isolation and differentiation of urinary microbes. It contains cystine and lactose and is electrolyte-deficient; the latter trait prevents the swarming of Proteus species. Cystine promotes the formation of cystine-dependent dwarf colonies. Bromothymol blue is the indicator used in the agar, it changes to yellow in case of acid production during fermentation of lactose or changes to deep blue in case of alkalinization. Principle:- De-Man Roagosa Sharpe agar is described by DeMan, Rogosa and Sharpe (1960). Media composed of proteose peptone, meat extract (equivalent to beef extract), yeast extract, dextrose, sorbitan monooleate, ammonium citrate, sodium acetate magnesium sulfate, manganese sulfate and potassium phosphate dibasic. Proteose peptone, meat extract and yeast extract provide nitrogen, carbon, vitamins and other necessary elements. Dextrose is carbohydrate source, Sorbitan monooleate, ammonium citrate, sodium acetate, magnesium sulfate and manganese sulfate are selective agents inhibits growth of other bacteria and essential for the growth of lactobacilli. Directions:- Dissolve 55.15 grams in 1000 ml distilled water. Boil De-Man Rogosa Sharpe Broth to dissolve the medium completely and sterilize by autoclaving at 15 lbs. pressure (121°C) for 15 min, cool it to 42-45 °C and inoculate test sample aseptically. Incubate in 5% CO2 atmosphere at 33-37°C for 24-72 hrs.

Principle And Interpretation

Haemolysins are exotoxins produced by bacteria that lyse red blood cells. The haemolytic reaction can be visualized on blood agar plates. On blood agar plates colonies of haemolytic bacteria may be surrounded by clear, colourless zone where the red blood cells have been lysed and the haemoglobin destroyed to a colourless compound. This is beta haemolysis. Other types of bacteria can reduce haemoglobin to methaemoglobin which produces a greenish zone around the colonies and is called alpha haemolysis.

Principle:- The presence of Bile salts, Sodium citrate and Brilliant green inhibit the gram positive bacteria most of coliforms and limiting Proteus spp. and allowing the Salmonella spp. to grow.
